How would it be explained that in the HDL test, the percentages of cytotoxicity are negative?
It is as if the cells treated with the drug the LDH released was lower than in normal cells
Percentage of cytotoxicity= Exp value - low control/high control - low control
Low control Provides information about the LDH activity released from the untreated normal cells (= spontaneous LDH release).
High control Provides information about the maximum releasable LDH activity in the cells (= maximum LDH release)
The most simple explanation is that the drug you are testing is a cytoprotective agent.
Yes ok, but although the values are lower than those obtained for cells without drugs, could one think of a cytoprotective effect?
Yes, even under basal conditions without any drug, cells are prone to undergo stress due to many reasons including rapid culture expansion that can cause proliferative stress, pH changes due to metabolites released during aerobic respiration and proliferation, generation of biological reactive intermediates as a product of normal cellular metabolism, excessive protein synthesis driven by expansion and the resultant accumulation of unfolded proteins and ER stress, etc. The cellular stress can lead to some damage to their cell membrane resulting in mild to moderate release of intracellular LDH even during growth in the absence of any exposure to extraneous drugs or chemicals. In such cases when the cells are treated with an antioxidant or inhibitors of cell death pathways or ER stress or even a suppressor of proliferation that can switch the status of cell cycle in rapidly expanding cells into quiescent stage, can all block the basal oxidative stress/ER stress/expansion-related stress that can salvage the dying cells and thereby protecting the cells under basal stress.
