Ideally, Cyclopamine treatment should not have any effect on expression of Hh gene itself. However, if Hh expression increases, it could be due to activation of a positive feedback loop upon inhibition of down stream signaling. Although, this is highly unlikely to be true in this case because Cyclopamine is being widely used to inhibit Hh pathway.
