Sonuji, you can use GC method or HPLC for quantifying the auxins content. For this you have use methanol or ethanol or alkaline water for extraction of auxins from the sample. but remember all the solvents you use should be of HPLC grades only. Please follow the attachment.

Also you should have the correponding standard.
