How hydrophilic compounds is your sample? Try using 2% acetic acid in methanol or reverse-phase TLC. Perhaps you can directly optimize in HPLC-reverse phase

It really depends on how hydrophilic your sample is. If you use the normal phase TLC, try optimizing your solvent system with 2% acetic acid in MeOH or you can go up until 5% acetic acid. Just make sure, the silica on TLC wont be dissolved.
