Hello Florian.. in this case actually i don't know well about Atlantic herring sperm. But in my experience about how to thawing bull frozen semen, the best method for thawing bull frozen semen is using water bath 30C during 30S for 0,5ml straw

We don't work with the same species but I suggest you to fixe post-taw semen and to evaluate cryo damage among different methods and combine your results with post-taw semen motility. It may give you some cues and help to explain your results. We work with Copper redhorse (Moxostoma hubbsi), an endangered species and made this kind of evaluation but all the results are not analyzed. As an exemple we use a 37-38 degrees bath for 8 seconds with 0.5 ml straws and about 20 seconds with 1.2 ml straws at the same temperature. I'm really interested in your results.

As for the herring you can try to use 10-20% DMSO after adjustments of sperm concentration to about 10^9 cells/ml (might be bit higher or lower, not sure about herring cells, but 10^9 is good starting point). Sperm concentration will be the key when use the DMSO. You can also try sucrose combined with methanol, however it will takes longer to establish exact final concentration of sucrose usefull for herring (might vary from 0.15 to 0.5 M), however the final concentration of methanol should not exceed 15% (10-15%) in this case.
