I have a novel species of bacteria which has the genes/enzymes responsible for carrying out methylotrophic activity. This strain can grow on methanol as well as methylamine (on the basis of presence of genes).
I want to physiologically invesitgate if it can grow solely on methylamine or not.
Can you provide the suggestions for testing this ability?
For methanol testing, I am using ammonium MineralSalts (AMS) Medium (ATCCMedium 784) and adding 0.5 g of methanol through filter.

Probably the easiest way to see if your methylotroph uses methylamine as a carbon source for growth is to just replace methanol with methylamine in the AMS media. I would prepare three flasks: one with methanol in AMS, one with methylamine in AMS; and one with no carbon added (AMS only) and follow OD as a function of time. Perhaps add methylamine at a few different concentrations as I am not sure of its toxicity. If you want to see if the organism uses it as both a C and N source, then you will need to prepare a medium without another source of nitrogen for growth (i.e., no ammonium or nitrate - which are common in media for methanotrophs). Prepare the N free medium and run the same basic treatments as described above plus one with methanol and ammonium added (as a positive control). The final option is to see if it uses methylamine as a N source only, in which case you would use the N-free medium for the previous studies and add both methanol and methylamine (as the only N source) and look for growth.
