We have cultured Fusarium oxysporum on PDA medium and we want to store it for a long period of time. We would be thankful if someone can provide an easy method to store fungal strain especially Fusarium oxysporum using sterile water.
We used lypholization to preserve important strains of pathogenic fungi, which cause disease in humans and animals.The lypholized strains of Cryptococcus neoformans and Cryptococcus gattii were kept for 15 years.You can try the lypholization method to preserve the culture of Fusarium oxysporum for long period.
Most fungi can be kept alive on many of the agar media for 6 months or 1 year before transfers are necessary. Sporulating agar cultures may be frozen at -20 C. in the deep freeze and maintained for considerable periods of time. Another good method for keeping cultures for a number of years is by pouring sterilized mineral oil in the test tube until the culture is completely submerged . The best method for preservation of most fungi that sporulate is by the lyophil process or the freeze dry method . Cultures have been maintained for at least 20 years by this method.(Alexopoulos and Beneke, 1962). you can also use sterilized sand for preserve of F.oxysporum.

You can keep the different fungi many many methods according to the period you wan. If you want keep the fungus for short period about 3-4 months, you can use suitable cultur media.
If you want to keep Fusarium oxysporum for a long time, you can use the dry plant roots which inoculated with fungus and incubated until the fungus well grows, then the roots with fungus kept under room temperature for a long time.
I use a simple method with water and glycerin. It can be stored in microtubes (2 mL) at room temperature for a long time without the necessity to transfer the culture periodically.
The formulation is simple: Water-glycerin 10%
Make the culture on PDA (or specific) agar media when it comes seven days of inoculation, cut small blocks of the culture, and then transfer the blocks to microtubes. (all the supplies must be previously sterilized)
Add 1,5mL of the solution water-glycerin 10% and storage.
The method can be used to storage filamentous fungi and yeast.

cryopreservation technique in liquid nitrogen is used for long term preservation: What I do is putting in cryotubes two or three discs of fungal culture and overlaying with glycerol 15%. And kept at -80% . Its really works. My collection of Fusarium dates from 2009!!!!
