Hi,
I am interested in staining frozen cochlear tissue with Massons trichrome stain for collagen. Following perfusiuon, the tissue was post fixed for 1 hr in 10% NBF, decalcified in EDTA, sucrose cryoprotected and finally embedded in OCT.
Many protocols perform this stain on paraffin embedded tissue, has anyone performed Masson's trichrome staining in frozen tissue?

Masson's trichrome is a routine stain on frozen sections of muscle biopsies that are part of daily diagnostic practice for neuropathologists. So, if you have any trouble with your stainings, contact your local clinical neuropath lab.
Thankyou everyone for our answers. Thanks Enrico Marani, we are going to incubate the sections in Bouin's fixative for 1hr at 60 deg C before staining. I will try this next week and see how it goes.
