I am trying to solubilize EGTA for an MES buffer I am making to perform cytoskeletal fractionation in endothelial cells. I am running into issues where the EGTA won't solubilize in water when I try to make a stock solution. So far, I have tried making 0.25M and decreasing the concentration, but I still find EGTA unsolubilized at the bottom of my tube. . I have seen that EGTA can be solubilized using Tris, but I'm concerned it will mess with other ingredients in my buffer. My buffer includes: 0.1 M MES, 2.5 mM EGTA, 0.5 mM MgCl2, and 0.1 mM EDTA. Does anyone know how I could solubilize EGTA either by Tris (knowing it won't interact with my other buffer ingredients) or with something else?
