I am conducting a wind tunnel experiment on NACA 4412. I use sandpapers with different grit sizes to simulate the roughness on the leading edge. I want to compare the experimental results with CFD simulation. But now I do not know how to simulate sandpapers in CFD?

To model sandpapers with different sizes you have simply to define surface roughness in FLUENT or any other flow solver. I advise you to use either Ansys CFX or Fluent. Nevertheless you may use any other flow solver.
