I am interested in obtaining the genomic DNA of C.elegans, but the outer cuticle layer reduces my total genomic DNA concentration, any suggestion on this?
Genomic DNA isolation from C. elegans is standard procedure. To assure tissue lysis it’s important to flash-freeze the worm pellet in liquid nitrogen before incubation in lysis buffer supplemented with proteinase K for 1-2 hours at 65 degrees (followed by inactivation of the proteinase K at 95 degrees for 30 minutes). If your yield is low you could increase the incubation time at 65. It's easy to find on line protocols for worm genomic DNA preparation, which outline these steps in great detail including the composition of the lysis buffer and the concentration of proteinase K.
