I am performing a research of propagation in chayote. Brief work: 1) sterilization of shoot collected from mature plant by 0.05% HgCl2; 2) Creation of callus from pieces of shoot/leaf (MS+0.5% sucrose+ 0.5-2.5 ppm BAP/BA+0.5-2.0ppm IAA/IBA/NAA); 3) Regeneration of invitro plant from sterilized mature shoots or callus (MS+ 0.5% sucrose + 0.5-2.5 TDZ/kinetin/BAP/BA/2iP/combination)
But the results in step 3 were very poor. The multiplication of shoot from sterilized mature shoots is very low and the new in vitro shoots were weak and die gradually. The inoculated callus on regeneration medium were browning and died after 3-4 weeks. There was no shoot regenerated form callus.
So, you experts could help me:
1) How to regenerate shoot from callus
2) Improvement the multiplication rate of in vitro shoot
3) Give advices something?
