Our project designed to distinguish different norepinephrine content in mouse liver between groups. Here come to our concerns. how to sac mice without disturbing their physiological sympathetic activity? Neck dislocation, CO2 Euthanasia or other methods?
Initial stages of methods such as gradual CO2 euthanasia can cause stress in the rodents and dramatically influence the sympathetic nervous system - see link 1 for more details. I would also consider corticosterone levels as discussed by H. Valentine in link 2. The stresses to the animals must be kept to a minimum to preserve physiological norepinephrine levels, and since overdosing by general anaesthetics would blunt the stress response lower than physiological levels (link 3), cervical dislocation appears to be the most appropriate method.
