Hi everyone,

I am recently focus on the recording of transient sodium current in the brain slice using outside-out model. But I have some issues that I don't know how to figure them out.

1. How can I tell the outside-out mode is formed? Are there any criteria?

2. I got a huge current when I step the Vm from -70 to -20 (holding at -70) labeled as "1" in the attached picture. I guess "2" represents the transient sodium, am I right?

3. What is the huge current? How can I get rid of this huge current during transient sodium current recording?

Here are the conditions that I used in the recording:

External solution: NaCl 106 mM, TEA-Cl 20 mM, KCl 3 mM, NaH2PO4.H2O 1.25 mM, CaCl2 1.6 mM, MgSO4.7H2O 1.5 mM, D-Glucose 10 mM

Internal solution: TEA-Cl included Cs+ based internal solution

Blockers: CdCl, 4-AP

Are there anything wrong?
