How to reconcile with downregulation of CD3, CD4 expression after PMA/Ionomycin stimulation?

Has anyone have experience with Down-regulation of CD3 and CD4 expression, when stimulating with PMA/Ionomycin and Brefeldin A?

Is it due to PMA/Ionomycin or due to the protein transport inhibitor Brefeldin A?

Is there a way around to cope with this problem such as lowering the PMA/Ionomycin concentration or shortening the time of incubation?

By the way I have been using 6h stimulation with 50 ng/ml (PMA) and 600 ng/ml (ionomycin).
