I have a weak culture of this sulfate reducing bacteria grown on aqueous medium recommended in DSMZ medium (low ionic strength medium) (25 degrees C), at very low OD value. Now, I would like to re-grow it in seawater to a higher cell count. May I know what is the best medium to regrow this strain in a short time? I will be grateful if you can share me your recipe. You want to try using the Lactate-Sulfate Media recipe described in some of Lee Krumholz's work (e.g. Li and Krumholz, 2007). I have had very good luck bringing up the OD for G20 using this recipe. You may even try to swap out the bicarbonate buffer with HEPES (want to adjust pH to about 7-7.5) and degas liquid and gas phase of your media prep with scrubbed (e.g. heated copper column) N2 gas prior to autoclaving.
