I grow this D. latus in syringe bottle under temperature 30 degree celsius incubator. I followed the instruction given by the DSMZ but it doesn't seem to work. The bacteria doesn't grow into high biomass. Attached is the pH and OD curve after inoculate the strain. I wonder if I missed something very essential here. If anyone could share experience in growing SRB (this particular strain) then it would be great. Many Thanks in advance.

Check the anaerobic status of your culture system and the medium before inoculation. You may try an anaerobic indicator like Methylene Blue, resazurin etc. for checking the anaerobic status of your culture medium. Best of luck.

I've put in resazurin and Na-sulfide. The resazurin turns colourless. I think the vial is in anaerobic condition but still can't get them grow properly.

Thank you for updating. It seems that your anaerobic culture system system is OK. As you know, anaerobic bacterial growth is very slow in general, and you can not expect a too high biomass as aerobes and But not sure of difference od new medium and new medium1. From the attached gragh, try to trnsfer an young & active inoculum cultrue of around 100h, to a new medium with a higher buffer capacity, if possible. Best luck.
