I have to conduct enzyme assay of around 200 samples to check their kinetic rate.
Traditionally, for small number of samples we carry out cuvette based spectrophotometry assay. But for such large number of samples carrying out individual spectro analysis will be time consuming and chances of error. It will be helpful if any alternative and parellel analysis technique/protocol suggested.
Do you have access to an absorbance plate reader? You could use one to make the measurements in 96-well or even 384-well plates. It is highly beneficial to also use a multichannel pipettor (8 or 12 channel) to put reagents into the wells. A plate shaker improves mixing of the samples in the wells. Thank you Adam for your answer. We do have microplate reader in our lab (Epoch2 and iMark). Although we currently dont have multichannel pipette that is what making this situation challenging. As additions carried out using single channel pipette wont be possible or reliable and correct when working on kinetics.
If you have large number of samples, take the absorbance in 96 well or 384 well plates, and also use a multichannel pipettor (as Dr. Adam said earlier). For all the above said enzymatic antioxidants such as catalase (CAT), ascorbate peroxidase (APX), and peroxidase (POX), take the absorbance measurements at 240, 290, and 420 nm. You can also check the kinetics of each enzyme.
