Dear all,
I am analysing some pictures of MCF10-A acini (3D), and one of the phenotypes observed is the formation of lumps, like lamellopodia features. I would like to quantify those lumps to see if this observation is really significant, but I am struggling with how I should do this in Fiji. I am trying to create an ROI and quantifying the area outside the ROI, but not all the acini are perfectly rounded or exactly the same, also, it seems that might be an easier way to do it. I uploaded one image of the lumps. Any suggestion is super welcome and thank you very much in advance!

Hi Camila de Castro Ferezin . The first step is thresholding. I just uploaded a video on counting and thresholding. Perhaps this will be useful to you. Area and sizing as well as other measurements will follow soon.
