I am working on encapsulating drug in Pre-PGS by solvent evaporation method. In which organic phase is PGS dissolved in THF, drug in water with tween 20. I am getting little particles at end. But my FTIR anlysis is showing hydrolysis of ester bond. No ester bond peak in final construct.
What should I do to prevent hydrolysis?
Curing PGS is not an option as it will make it insoluble...........
Dear all, your actual problem occurs while doing thermal evaporation. You can evaporate under reduced pressure at the lowest possible température, but the best will be freeze-drying. My Regards

Thanks Abdelkader BOUAZIZ ... you mean its getting hydrolysed during evaporation stage..not during mixing......but i am not giving any temprature, I am evaporating it on room temperature under continuous stirring.... should I go for rotary evaporation??? Or could the probe sonication be the reason? Though i perform sonication for mixing both phases upon ice.....
Could pH be the factor.. its 5.5....... may be due to tween???
How much time does it take to evaporate at room temperature? This time if too long it adds in favor of hydrolysis. Yes, rotavap is to be used at reduced pressure and low bath temperature. Best of Luck
Too long if it is to avoid or reduce hydrolysis. Try the rotavap. Good Luck

The conversion degree of carboxylic acid and the esterification degree depend on the temperature and the reaction time as well as the glycerol/sebacate ratio.
