Dear all,
I cultured C. difficile into BHI under anaerobic condition and when the cells reached the mid log phase, I centrifuged them and washed them twice with pre-reduced PBS (pH 7.4). Cells were placed under anaerobic condition and measured four times in one hour as a negative control after addition of only pre-reduced PBS. Unfortunately, the absorbance of the cells at OD600 decreased through the time from 1 to 0.5 -0.8 . Would any suggest what could be wrong in my procedure, please?
