I'm growing mucuna seeds invitro. I soaked mucuna seeds in 1% ascorbic acid for 3 hours and then inoculated in basal media with 1 mg/L GA3. Though it's germinating, media is turning black. Should I worry about it? Or just ignore if plants continue to grow?
add PVP in the media and see in which concentration blackening stop as well as plants grows well.
Turmeric (cucurma longa) has been used as an antioxidant agent in medical as well as health sciences some years ago. However, it has not been applied in tissue culture until now. Browning tissues is a result of tissue damages caused by polyphenol oxidase enzyme. In order to study this problem and reduce browning, explants are cultured in culture media containing antioxidants including citric acid, ascorbic acid 1,4 dithiothreitol, mercapto ethanol.
And also charcoal. Washed one. In order not to change the pH of the medium. It is always written to use 0.5%. But I use 0.02 %. 0.1 % already makes the medium "black". And could absorbe too much of the nutrion.
If don´t it affect germination, don´t add nothing. In Phaseolus vulgaris that happen without problems on germination.
Agree with Luis Pinzón . If germination is not affecting. leave it as such. however if affecting anything suggested above can be added in media.
