in order to conjugate an antibody to a cytotoxic drug, I wan to use SMCC linker. I have a two step reaction. in the first step i want to conjugate SMCC linker to the antibody and in the second step conjugate drug to antibody-linker. SMCC is dissolved in DMSO. when I add linker solution to antibody solution, antibodies aggregate. DMSO is 5% v/v. I added the DMSO without linker and aggregation was also occurred but the aggregates were smaller. how can I prevent antibody aggregation during reaction?
