I need to add ascorbic acid into solid medium. I prepared the medium, adjusted pH to 5.4 (pH for ND medium), autoclaved it, added ascorbic acid (0.58 mM; sterilized by syringe filter), and then poured on Petri dish, but I've got a problem that is ascorbic acid decrease pH. I would like to know...
Should I concern about it?
Does anyone know the way to prepare the medium containing Ascorbic acid without pH alteration?

I guess, this depends on what your project is about:
since it's called ascorbic ACID the sour part may be part of the effect you're trying to achieve. If this isn't the case, one solution could be, adjusting two media, one at 5.4pH, one at a higher pH, that results in 5.4, when ACA is added.

Calculate the relative amounts of ascorbic acid and sodium ascorbate (using Henderson-Hasselbach equation) that gives a pH of 5.40. Make that buffer solution. That is what you will add to the medium, keeping its pH at 5.40.
