I want to do a comparative analysis of wild plant vs callus through LC-MS. I have prepared plant extract in methanol 1 gram plant powder in 10ml methanol. Now I want to remove chlorophyll contents using petroleum ether. How its done can anyone help me?? How much and For how long petroleum ether should be mixed with methanolic extract. I don't want anything else be removed from extract except chlorophyll.
Best option; Chlorophyll A-B binding protein as immunoaffinity removel this will present the most effective elimination and would be completely non-invasive to your metabolites.
Secondly, due to the central magnesium, you may pass the organic extract through EDTA-loaded gravity flow SPE (Chelex in replacement) to perform Mg chelation, thus Chlorophyll removal. High specificity, and most of the metabolites will remain in the extract. The batch-dispersive mode can also be used.
Thirdly, they can be centrifuged out. Chlorophyll micelles form in ethanol if you introduce a little water.
Why do you need to? Removing chlorophyll with solvent will remove other metabolites as well. Chlorophyll will be present in different amounts but you can identify it by LCMS (easy if you have PDA/UV as well) and just disregard it from subsequent analysis. Depends how much your wild plant has maybe?
