Dear Zaheer, usually ICPI is done for NDV while pathogenicity for AIVs is checked by IVPI that is performed on 4-6 weeks old chicken.
Traditional pathogenicity tests like ICPI, IVPI and mean death time in embryos can all be used for both NDV and avian influenza. I want to know the place and depth of injection.

We are doing this test regulairly for ascertainig fremedom from AE virus contamiation. The assistant presents the chick to operator while the superior positiion of skull is approachable. draw an imaginary line dividing skull in two equal halves.few milimeter lateral to midline at the level of ocular orbit enter the skull by sharp needle 1--2mm inside. 100 microliter syringe fitted with .5cm needle 20 gauge is adequate. injection is usually uneventful. some times chick may show little excitation but soon settle. very rearely chick may die. we have to use one more chick discarding accidental death. shivdekar

The ICPI is done for testing pathogenicity of NDV. The test is performed on 1 day old chickens (between 24-48 hours). Pathogenicity for AIV is checked by IVPI and the test is performed on 4-6 weeks old chickens.

Hello, ICPI is so far the most accurate in-vivo biological test to as certain pathogenicity of NDV strains. I have done several of such in my studies. Many publications show out the procedure well but you need to understand that some key steps are left out.
