I tried to prepare complex inclusion of essential oil in alpha and beta cylodextrin.
I mix Essential oil + cyclodextrin (15mM:15mM), I filter them 0.45µm then I freeze dried.
I solubilize the obtained power in ethanol and I use spectroUV to measure.
However, I have problem how to determine the 100% of essential oil.
the results that I obtained showed that my 100% essential oil is lower than the encapsulated essential oil which is not logic.
Have you an idea what is the mistake that I did?

Is your essential oil volatile? Or, are elements of the oil volatile?
With a low enough pressure and enough temperauture almost anything can sublime / evaporate. Depending on the preparation process there may be a small % water in the sample, or other light element, that evaporates.
Suggest analysing the condensate in the cold trap.
If you have it available, try NMR before and after.

The lambda max may shift after inclusion in cyclodextrin. Use HPLC for separation or prepare a standard with the same ratio as your sample.
I feel there is a mistake in extracting the total oil from the encapsulated powder. Also quantify with HPLC at least to believe your results.
