Hi, I want to know how I can measure the concentration of sulfate and iron of the anaerobic sludge accurately? Generally I use atomic absorption spectroscopy (AAS) for total iron measurement and turbidimetric method for sulfate, but for sludge particles what should I do for sample preparation? Do I need to acidify the sample and digest it?

Typically membrane filtration is used to get rid of fine particles and microbes. You may check https://www.sartorius.com/sartorius/en/EUR/products/laboratory/filters/syringe_filters but there is a lot of companies that offer similar solutions. Further you may try centrifugation first, which also would prevent or delay filter clogging.
Digestion would dissolve Fe and S bound to the particles and possibly would alter SO4 by some redox processes, which possibly you don't want to happen. So if you need dissolved Fe and sulfate you should not digest your sample.
