I am preparing fluorescent MIPs however how do i measure their fluorescence on a spectrofluorimetre while they are suspended in the solution as solid. As far as i know all fluorescence measurements have to be carried out using a clear solution or the particles would impede the light from the excitation source.
Anyone has an idea how that works?
The molecularly imprinted polymers (MIPs). MIPs demonstrate a wide range of applicability, good plasticity, stability, and high selectivity, and their internal recognition sites can be selectively combined with template molecules to achieve selective recognition. A molecularly imprinted fluorescence sensor (MIFs) incorporates fluorescent materials (fluorescein or fluorescent nanoparticles) into a molecularly imprinted polymer synthesis system and transforms the binding sites between target molecules and molecularly imprinted materials into readable fluorescence signals. This sensor demonstrates the advantages of high sensitivity and selectivity of fluorescence detection.

You are right. Fluorimeters are designed to work on solutions with low absorbance.
You work on solids.
If the solid can be dispersed into a fine powder into a solvent, you can study these suspensions with a spectrofluorimeter.
But take care : a fine enough powder is composed of particules small enougth so that absorption is proportional to depth inside the particle. The scattering of the suspension in the cuvette has also to be small.
If you work with dense highly scarttering solids, you should study the back scattered light from your samples and apply the Kubelka Munk theory.

Well that depends on which spectrofluorimeter you have access to. For example, I use an ISS PC and I can put in a variable angle cuvette holder to allow me to use a front face arrangement to look at the fluorescence from solid surfaces. I have attached a picture of this form my book.
Prepare a diluted suspension of your MIP, centrifuge well and decant to get a clear solution. It will work in a good way.
Hellow Dr. Azza and Dr. Aya I think that high fluorescence when the solution is diluted and with low temperature, also additon of fluoregenic reagent can increase the fluorescence intensity and choosing the suitable solvent. Azza H. Rageh and Aya M. Mostafa
