Il have a same answer. But u can also use ethanol instead of butanol for preparing solution. I prepare without acetic acid than also the charring pattern was good.
I've used ninhydrin solution to stain amino-phospholipids in TLC plates. The staining solution was prepared by weighing 0.2gr in 100mL of butanol saturated with water (vortex butanol in water and collect the lower phase). Hope this helps,
There are a few different versions of the recipe for ninhydrin stain solution but most include a little acetic acid, such as this one:
0.2 g Ninhydrin
0.5 ml Acetic acid
100 ml n-Butanol
4.5 ml Water
