I`ve been attempting to make minimal medium with the following components:
0.5 g/liter asparagine,
1 g/literKH2PO4,
2.5 g/liter Na2HPO4,
50 mg/liter ferric ammonium citrate,
0.5 g/liter MgSO47H2O,
0.5 mg/liter CaCl2, and
0.1 mg/liter ZnSO4
Can someone please advise on the components that should be filter sterilized/which can be autoclaved and the order of adding components as I`ve tried autoclaving all together but a precipitate formed
I was of the understanding that a minimal medium is composed strictly of inorganic compounds. Addition of asparagine to the medium, no matter how small, disqualifies the medium from being a basal one. Secondly, the ferric salt used could be the source of the precipitation except you specifically require that salt in the medium. for a minimal medium, ferric chloride or ferrous sulfate are preferred. Like Martin said, it could be a function of the pH. The ratio of the dibasic to monobasic potassium phosphates is a bit high and could be responsible for the precipitation.
