I have been trying to produce GUVs via electroformation.

I use a 2mM DOPC (+ 1% rhodmine B - PE) chloroform solution that I spread on ITO coated glass slide and put in a dessicator for 2h. I make a chamber by making a 1mm thick pdms membrane that I clamp between the glass slides. Once the chamber is ready I slowly add a 10mM Tris buffer pH:7.5.

Then I apply a voltage of 2V for 2h at 10Hz. Then I decreased the frequency to 5Hz for half an hour. Finally I take the solution out the chamber with a needle and put them into a cell counting chamber. I can see that there are some small GUVs formed 1-5 µm. But which parameters should I change to get bigger GUVs?

(Sidenote: is there somebody who nows a better way of imaging the GUVs, I have the feeling the cell counting chamber is not ideal)
