Hey everyone! I have a naive question about SeaHorse analyzer, maybe some of you faced with this problem. In the lab we tried to adapt the SeaHorse mitochondrial stress protocol using bulk metabolic inhibitors from Sigma. At the beginning everything worked totally fine, but later on we found out that our FCCP stock had lost its activity just in a couple of weeks after reconstitution. We tried different approaches like dissolving it in 96% ethanol or DMSO, storing at -80, nothing helps. All the other reagents are stored pretty well. Actually, this is the first time I meet the molecule that is so unstable in standard solvents stored at -80. Does anyone of you dealt with such a problem and found a solution or the only way is to purchase Agilent kits for mitochondria stress test? Weighting microgram amounts for each experiment is also an option, but we don't have the scales with such precision..
