in our lab we prepare the PFA as follow:

weight 4g of PFA (powder) every 100 ml of 1X PBS (we have tablets of PBS to dissolve in water to prepare it).

After adding the PBS, we add 50 ul of NaOH 10M (it won't dissolve otherwhise).

We then leave the solution at 60 C (with a stirrer or with agitation in a water bath if possible) until it's clear.

You can then check the pH of the solution (should be 7.4).

Then, you can either put it in the fridge or on ice until it's cold or aliquot and keep it at -20 until you need it.

Hi Shahid, we use a method similar to what Valeria described:

For 50 ml of 4% PFA weigh 2 mg of PFA powder into a 50 ml conical tube. Add 20 ml of Di water and 100 ul of 1M NaOH. Put the tube into an end-over-end mixer overnight at RT. The PFA will be dissolved.

Then add 5 ml of 10X PBS and fill to 50 ml.
