Hello people! I have some gilled mushroom spore prints(on aluminum foil) one with 4 years and I need to have sure with they come to germinate on my culture media (PDA) or on my wheat spawn. What kind of test I need to make to measure the spores viability to not waste the materials here?
The obvious answer is to periodically take a small subsample, plate it on PDA and count the % germination of spores under magnification. Since that's obvious, I think you must be wondering if you can use some other method? If I'm not understanding you correctly, perhaps you could elaborate a little.
Sorry for misunderstand. But I think on a microscopic method maybe with reagent to look some spores under magnification and see some viability or no from the structure of spore. Here don't have much resource to spend agar and dextrose, so I think on easy methods for this verification
