I do not have an anaerobic chamber in our lab but I want to isolate the anaerobic bacterias from a glycerol stock. So, is it necessary to have an anaerobic chamber for isolating anaerobic bacterias? If no then what are the procedures available for isolating anaerobic bacterias from glycerol stock without an anaerobic chamber?
The are some old but solid tests. Hungate roll tube- but this relies on specific types of tubes. More available in a general lab is deep agar overlays. You mix dilutions of your glycerol stock suspension in an appropriate agar medium, mix and let solidify in a headspace of oxygen-free nitrogen. When solid, overlay with molten paraffin wax, which also solidifies. This forms a barrier to oxygen.

And bacteria is already plural, singular would be bacterium.
