Hello Ibomcha Ngangom .Particular gene it depends on ur gene of interest. for eg if u r int in protein ferrititn function search for previous study (literature). browse for sequence available in data bank for eg (NCBI) find the sequence for the gene which codes for ferritin. then design a primer for ur gene. go for PCR.
After PCR..perform AGE - and compare size with markers..then transform into E. coli or suitable hosts..perform PAGE and look for any different proteins not commonly found in E. coli
if you have the protein sequence then by going reverse you can obtain the nucleotide sequence of the gene. this can be done by bioinformatics softwares. design primers for that sequence and run PCR.
