Anther derived plants of oil seed(niger) were raised in-vitro. After successful rooting, hardening is becoming difficult . Can any one suggest for proper hardening conditions for 100% plant survival.
Harden seedlings in an unheated greenhouse or cold frame. Place seedlings and plants into the structure for a couple of hours on the first day, then gradually increase the length of time they are in place by two or more hours per day :)
doubled haploid plants are weak ( and much more the haploid ones).
achieving 100% survival may be too ambitious a goal. I would advise you to multiply your lines before rooting them, so as to increase the chances of having at least one plant of each pure lines. A method of acclimatization could be hardening already in vitro: there are containers such as those of the Duchefa that have filters for gaseous exchanges. Using them in the rooting phase,you can obtain a first hardening of the seedling. Then, transfer to the greenhouse, in a substrate with 50% agri-perlite or sand and 50% substrate for cultivation, fertilizing perhaps with only half-strengh solution of MS, use the fog under small tunnels made with polyethylene sheets and, if the light is too much, with shaded nets (about 60%) , after the first 3-4 days in conditions of high humidity, adjust the fog to give less water and, a few days later, open the small tunnels a little and decrease the shading and go on in this way until to completely remove the fog and shading and polyethylene sheets after 15-20-30 days. You have to experience your own species.
It might be worth trying antiperspirants like pinolene as well, but I have no experience with it.
