Dear all,

I am struggling with CK666 inhibitor which is not taking effect in my experiment. Here is my system :

- I have MII Mouse oocytes in a 2uL AKSOM (media)+100 uM CK666 droplet covered with 180uL Lightoil (Mineral oil) in a well of a 4well 35mm glass bottom dish.

The expected effect should be the disassembly of the actin cap of the MII mouse oocytes. After 3 hours of live imaging (Actin labelled with UtrCH-GFP), I still see the cap.

Here, I have the feeling that my drug completely diffused out to the oil. I used other drugs before but it is not as dramatic as CK666.

If you please have any experience with CK666 and how to use it in these cases would be very helpful.

I m thinking in adding the drug into the oil but i do not know how soluble that is. Same if I go for higher concentration in my droplet then how much will diffuse out and how fast.

Your recommendation would be much appreciated.
