I am working in fungal ( trichoderma ) batch cultivation with rice & soya flour after inculation culture grown fine but not sporulated .
how to grow fungal sporulation in shake flask and fermenter
please give me valuable comments
Lala K Nagaraj Trichoderma generally sporulates on Petri plates and on suspension (Potato Dextrose Broth - that I have worked with). Generally, a fully grown plate of Trichoderma is used - add 4/5 bits per flask i.e. generally 200 ml of broth keep it in a shaker for 6/7 days to achieve spore suspension. If you need a specific medium you may replace the potato with your media and give it a try. pH should be 6.5-6.8.
Jiwan Paudel , hi sir thanks for your suggestion could you please tell shaker RPM and temperature , i am using normal conical flask , in 200 ml broth shall i use 500ml conical flask
please response me sir
Lala K Nagaraj Shaker speed can be maintained at 160 to 180 rpm and temperature can range from 26 to 28 degrees Celsius. You can add 200 ml broth to a 500 ml conical vessel. All the best.
