Hey all, I've been trying to grow fresh isolated mouse hepatocytes on collagen type I coated coverslips for fluorescence imaging in 6 well plate. Cells don't attach. Before that I've using pre-coated plate, cells were attached just fine, making a nice monolayer. Are there any special tricks? I've been using coverslips form neuvitro.
It is depending on the collagen solution (salt concentration etc.) and protocol (how the cover-slips were prepared and time to wait the coating solution to dry, etc.). I would try to trouble shoot first to use the collagen coating solution and protocol to coat a non-treated plate and compare the cell growth with or without the treatment to those precoated plates. If it is still have problem, I would by the pre-coated slips.... here is website has it.

Ching, thanks for your comment. I'm actually using this pre-coated coverslips from neuvitro and the hepatocytes haven't been attached. Before that I've been using pre-coated plates from Millipore, cells were growing just fine. I contact the neuvitro, they suggested to try to coat the coverslips with the collagen..

It will grow normally but the key of growing all types of cells is the type of cultured media, so if you use the suitable media you could change the substrate you culture on it as you prefer
