I want to make my own yeast extract from saccharomyces cerevisae for microbiological media. I need a exact amount of yeast, so wanted to know, how much grams (molar mass is preferred) of glucose/saccharose I will need to make 2 grams of yeast out of 1g starting culture.
I meant doing it that way, because else I would have to precipetate, filter, freeze dry andund weight the product, which is inconvenient and expensive.

Normally, yields of 0.45-0.50 g per g glucose on dry basis at the maximum specific growth rate can be reached but optimal other conditions. It means that the onversion rate is about 50%.

Baker's yeast is grown under fed-batch conditions. Here the yeast will utilize the dissolved oxygen at a very fast rate and then if excess of glucose is present, it will take to anaerobic fermentation. This is the reason why both dissolved oxygen and glucose concentration is monitored so that the yeast do not take to anaerobic fermentation and at the same time the organism will not be under stress.
