I am working in drought and salt stress with Arabidopsis and Tomato. I need to give these stress conditions to minimum 2 week old seedling.
Could I give these stress treatment in soil by giving equal volume of PEG or sorbitol solution (drought) and NaCl solution (salt) and water (Control)?
I am more doubtful in case of drought treatment instead of giving dehydration treatment could I follow sorbitol treatment in soil?
Drought should be in soil, using dehydration is not drought. If plants are of different size, then the bigger one will lose water faster and will show wilting- so you must make the test independent of plant size. This can be done using a gravimetric approach of weighing the pot and keeping the drought level constant.

Please do not use PEG or sorbitol. They induce a very fast shoot dehydration that does not happen in nature, In fact, they induce a strong decrease of Fv/Fm parameter (revealing damage of photosystem II), which does not happen in real drought.
It's better to apply drought by water withdrawal (partial or total). It's very important to have the same amount of soil in every pot at the beginning of the experiment, for comparing pots and for reproducibility. Then you can adjust the rate of soil dehydration by testing different pot sizes.

Hi gayathri, giving stress to plants in soil will not be appropriate stress condition. Since you want to give stress for two week old plants its better to go with hydroponic condition than soil treatment. In case of hydroponics roots will able to under go proper stress conditions comparative to soil treatments. In case of drought stress you can use mannitol for Arabidopsis. For arabidopsis salt and drought stress you can go through my recent publication "Expression of OsWNK9 in Arabidopsis conferred tolerance to salt and drought stress" in plant science journal, you can find all relevant experiments from that paper. Also for arabidopsis hydroponics i have attached the pic, you can go through that procedure for arabisposis hydroponics.

For gene expression pattern analysis at different hour stress treatment, people are suggesting to do hydroponics. Like for drought treatment, either giving PEG/sorbitol/mannitol or dehydrating the plants and NaCl solution for salt treatment. For phenotyping analysis, people do in soil either giving PEG/sorbitol/mannitol or water withdrawal for drought treatment and NaCl solution for salt treatment.
I am till now following hydroponics which I feel better for my experimental analysis.
