I am trying to get our new LC MS/MS set up to analyze triglycerides and am a little unsure about the proper solvents to use in order to get the best visualization. As it stands now I am going to use 5mM Ammonium Formate buffer in HPLC water and Acetonitrile with a RP-18 column. I'm not too concerned with the gradient time or run length as all that can be optimized but am curious if I am on the right path as far as solvents are concerned.
Noel is totally correct. 100% ACN is not strong enough for eluting most TAGs. The most common solvent for this purpose is IPA, which is miscible with water and ACN. There are so many papers on TAG analysis by LC-MS, which give you the detailed mobile phase compositions.
Noel W Davies Thank you very much for your response. I am confused what you mean by a mixture of Hexane and ACN however, are you referring to an isocratic gradient? Unfortunatly I don't have the means to see most the papers on LCMS TAG analysis since I can't purchase any papers.
