I am working on a prostate cancer mouse model with PC3 cells in nude mice. Does anyone have experience with the best number of cells for inoculation and if addition of matrigel is necessary?
Yes, I believe nude mice should work (see here: http://altogenlabs.com/xenograft-models/prostate-cancer-xenograft/pc-3-xenograft-model/). There are a few factors to consider when making the final decision about the protocol; eventual tumor growth or metastasis are dependent on the goals of given research. Matrigel is usually a good addition to the initial injection, as it helps with tumor formation.
Hello, we have had luck implanting PC-3 cells at a number ranging from 2-10 x10^6 in a 50% matrigel:PBS suspension of cells. We do however utilize male NSG mice in an effort to increase take rates.
