تست TTC برای تشخیص زیوایی بذر (مثال :بادام وحشی، همین روش با اندکی تغییرات برای بذرهای دیگر هم مورد استفاده است)

1-انتخاب 200 بذر به صورت تصادفی

2-شستشو و ضد عفونی بذرها با محلول هیپوکلریت سدیم 3% و سپس محلول قارچ کش بنومیل 3 در هزار

3-قرار دادن بذرها به مدت 24 ساعت در آب مقطر

4-حذف اندوکارپ و سپس پوشش های اضافی بذر و پوشش نازک قهوه ای روی بذر

5-تقسیم بذرها در 4 پتری دیش به قطر 11 سانتی متر در بین دو لایه کاعذ صافی

6-افزودن 6 میلی لیتر محلول TTC 1% و قرار دادن بذرها در ژرمیناتور در دمای 25-20 و رطوبت نسبی 80% در تاریکی

7-پس از 12 ساعت بذرها در زیر لوپ بررسی می شوند تا مشخص شود که بخش های مختلف بذر شامل لپه و رویان تغییر رنگ داده و به رنگ صورتی درآمده اند یا نه؟ رنگ صورتی نشان دهنده ی زیوایی بذر است

thank you for your comments. I have been writing all my answers and comments in English but in this case the question related to an Iranian (Persian language) colleague. for this reason i wrote my comments in Persian. Sorry for that.

Dear Nadine this the translation of my comments

1-Pick 200 seeds randomly.

2-Cleaning and disinfection of seeds with 3% sodium hypochlorite solution and fungicide Benomyl 3/1000.

3-Soaking the seeds in DW for 24 hours.

4-Remove endocarp, additional cover and yellow-brown thin coating of seeds.

5-Divide the seeds in about 4 petri dish carpeted with wet filter paper.

6-Add 6 ml solution of TTC 1% and put the seeds in the germinator at 25-30 C and 80% relative humidity in the dark.

7-After 12 hours the seeds are examined by a loop to determine the color change of the different parts of the seeds such as cotyledons and embryo (become pink or not)? the rate of pink color represents seed viability and germination.
