I want to determine degree of sulfatation, fine structure, monosaccharide composition, and molecular weight of fucoidan. But I don’t know exactly what method is needed to do these things (eg,what column of HPLC or ion exchange chromatography is appropriate for fucoidan purification )
Sulfated polysaccharides from marine algae are one of the commercially beneficial compounds with a range of pharmaceutical and biomedical applications. Sulfated polysaccharide are isolated from Sargassum myriocystum whichis identified as Fucoidan. Infrared spectra can be recorded on a MAGNA-IR750 Fourier transform spectrometer (INCOLET INSTRVMENTW, USA). For the spectrum range from 1500~2200 cm−1, 32 scans were collected at a spectral resolution of 4 cm−1. Pure fucoidanase (10 mg) is mixed with 100 mg of dried potassium bromide (KBr). Water vapor is purged from sample room. The spectrum of the amide I band of fucoidanase is obtained, and self-deconvolution and curve-fitting methods is used to analyze the secondary structure of the fucoidanase. The total neutral carbohydrate of the purified fucoidan is determined by the phenol-sulfuric acid method at 490 nm (Dubois et al. 1956) using L-fucose as a reference. Saccharides of fucoidans is performed by HPAEC (High Performance Anion-Exchange Chromatography) as described by Lee
et al. (2006), using Bio-LC (DX 500 Chromatography System, Dionex Co., USA) system equipped with a pulsed amperometric detector (ED 50, Dionex Co., USA)Infrared spectra is recorded on a MAGNA-IR750 Fourier transform spectrometer (INCOLET INSTRVMENTW, USA).
