Good morning everybody, We are piloting an "avatar" tumor injection model, which consists of MDA-MB-231 tumor cells in Matrigel inject in NOD/SCID mice. I would like to know if anyone has already frozen these samples in OTC, because we went to make hystological secctions in a cryostat and the sample is cracked, we are unable to make slides from that samples.

Here are some tips from others who may have experience with freezing MDA-MB-231 tumors in OCT:
- Pre-Cooling: Some researchers pre-cool their samples on ice before embedding them in OCT to slow down the freezing process.
- Additives: In some cases, adding substances like DMSO or sucrose to the OCT can help to prevent ice crystal formation.
- Sectioning Temperature: Adjust the temperature of the cryostat stage to find the optimal temperature for your samples. This can sometimes be a few degrees below the freezing point of OCT.
