After reaction, the mixture is added to cuvete and uv spectra performed

you should use a negative control (water or solution you make dilution), and compare with nanoparticles and drug. If you find some shift mean the particle react with drug.

A lots of thanks, by this way it is very easy, but any other way regarding any calculation and drug interaction and release process. If possible please suggest.

I am not sure about this. You need to perturb the interaction to comfirm it. If it is electrostatic add some salt and you should see release of the drug, if it is hydrophobic add some organic solvent or a similar compatible molecule.
