Sir, could you plz tell me how to find immobilized protein on the material and how much enzyme is needed to do soluble enzyme assay (trypsin) using BAPNA as substrate.

For example, I have prepared (1mg/ml) trypsin solution(10 mg trypsin in 10 ml buffer solution), and O.D. was reported before immobilization is 0.148 & after immobilization, I have got O.D. in the supernatant is 0.025.

Now, for solubilized enzyme assay, I couldn't able to find how much enzyme gets immobilized on the material and what amount of enzyme I have to attack for doing soluble enzyme assay.

Plz, help me so that I can be struck out of this situation...
